چکیده

Background: Mast cells (MCs) are essential in innate and adaptive immunity; however, strict activation control is critical to prevent unwanted inflammation. The demand for MC regulation especially important at the skin interface, where MCs remove pathogens but should not react commensal bacteria (CB). This study demonstrates that, microenvironment, mast assume a tolerant phenotype reduce unnecessary inflammation contact with beneficial CB via crosstalk dermal fibroblasts. We identified unique human (hMC) pathways modulated by fibroblasts (dFBs) using RNA sequence (RNAseq) single-cell (ScRNAseq). Results: increased communication between hMCs dFBs induces dramatic changes dFB populations hMC differentiation. ScRNAseq revealed that general direction co-cultured lose immune activation-related genes, making less 'active' narrowing or fine-tuning of To further understand how tolerance established presence dFBs, we performed an RNA-Seq stimulated gram-positive supernatant absence dFB. week dFB, separated them, challenged them CB. combined analysis confirmed activates inhibitory pathway NF-kB. hyaluronic acid (HA), extracellular matrix product, as mediator responsible regulatory zinc finger (de)ubiquitinating enzyme A20/tumor necrosis factor α-induced protein 3 (TNFAIP3), reduced response. Conclusion: Tolerant dermis maintain discovery HA tolerogenic ligand on opens new avenues treating inflammatory disorders.

Extracellular superoxide dismutase ameliorates house dust mite-induced atopic dermatitis-like skin inflammation and inhibits mast cell activation in mice.

Extracellular superoxide dismutase (EC-SOD) is an enzyme that catalyses the dismutation of superoxide anions. It has multiple functions, such as reactive oxygen species scavenging, anti-angiogenic, anti-inflammatory, antichemotatic and antitumor activities. Recently, we demonstrated that EC-SOD inhibits ovalbumin-induced allergic airway inflammation in mice. However, the anti-allergic effect of...

Angiopoietin1 Inhibits Mast Cell Activation and Protects against Anaphylaxis

Since morbidity and mortality rates of anaphylaxis diseases have been increasing year by year, how to prevent and manage these diseases effectively has become an important issue. Mast cells play a central regulatory role in allergic diseases. Angiopoietin1 (Ang-1) exhibits anti-inflammatory properties by inhibiting vascular permeability, leukocyte migration and cytokine production. However, Ang...

Stress-Induced Mast Cell Activation in Glabrous and Hairy Skin

Mast cells play a key role in modulation of stress-induced cutaneous inflammation. In this study we investigate the impact of repeated exposure to stress on mast cell degranulation, in both hairy and glabrous skin. Adult male Wistar rats were randomly divided into four groups: Stress 1 day (n = 8), Stress 10 days (n = 7), Stress 21 days (n = 6), and Control (n = 8). Rats in the stress groups we...
